Antibody binding efficiency depends on antibody origin and subtype. For antibodies with low Protein A affinity, extend incubation time (30-120 minutes), increase binding buffer pH (8-9), reduce ionic strength (25-100 mM NaCl), or select higher affinity ligands like Protein G or Protein A/G.
Store beads at 2-8°C and avoid contamination or drying. Aggregation in low pH elution buffer is normal and doesn't affect performance. Add 0.1% nonionic detergent (NP-40, Tween-20, Triton X-100) to buffers to prevent aggregation. For aggregated beads, wash to neutral pH and use 2-minute ultrasonic water bath to restore homogeneity.
