What is Sennosides?
Sennosides are anthraquinone glycosides extracted from the leaves of the senna plant (a legume), primarily comprising sennoside A and B, among other active ingredients. Their molecular structure consists of anthraquinone aglycones and multiple glycosyl groups, exhibiting good water solubility. As a core component of traditional stimulant laxatives, sennosides exert their laxative effect through intestinal metabolism, but strict control of dosage and duration of use is necessary.
Feature
Stimulating intestinal motility and acting as a laxative: Sennosides are not absorbed in the small intestine. After entering the colon, they are hydrolyzed by bacterial enzymes into active aglycones, which directly stimulate the sensory nerve plexus of the colonic mucosa. It enhances intestinal propulsive peristalsis and inhibits water absorption, typically producing a mild laxative effect 6-12 hours after administration.
Promoting electrolyte and water secretion: The active aglycone inhibits the activity of Na⁺-K⁺-ATPase in intestinal epithelial cells, reducing sodium ion absorption and promoting active chloride ion secretion. Increased osmotic pressure in the intestine leads to significant water retention, softening and increasing stool volume, further stimulating the defecation reflex.
Potential anti-inflammatory and antibacterial activities: Its anthraquinone structure has an inhibitory effect on some pathogenic bacteria in the intestine (such as Escherichia coli), potentially regulating the balance of intestinal flora. Low doses have a mild alleviating effect on local intestinal inflammation, but high doses may induce an inflammatory response.
Applications
Short-term treatment of functional constipation: As a component of prescription or over-the-counter laxatives, used for acute constipation or bowel preparation before bowel examinations. Suitable as a short-term alternative when other mild laxatives are ineffective, but continuous use should generally not exceed 1-2 weeks.
Special medical applications: Used after anorectal surgery to soften stool and reduce wound tension during defecation. Used to prevent fecal impaction in long-term bedridden patients, but should be used cautiously under the evaluation of a physician.
Traditional medicines and compound preparations: As a component of traditional Chinese medicine formulas for "purging heat and stagnation" (such as modified Da Chai Hu Tang), used for the treatment of constipation due to excess heat. In modern compound laxatives, it is often used as a fast-acting component in combination with bulk-forming laxatives.
