Testing Principle
Toluidine blue has strong heterochromaticity. When acidic mucopolysaccharides are present in the cells, especially sulfur-containing acidic mucous substances, the color of toluidine blue can be changed from blue to red. The basophilic particles in basophils are mainly acid mucopolysaccharides and belong to sulfate esters, which are heterochromatic. Therefore, staining with metachromatic dye toluidine blue can make them appear heterochromatic red-purple.
Stain Results
The basophilic granules are red, light pink or purple, and the background cells are light blue.
