Testing Principle
After staining acid-fast bacteria with the fluorescent dye Auramine O, the observation under a fluorescence microscope equipped with a violet-blue excitation filter (wavelength: 350–490 nm) reveals bright orange-yellow or yellow-green fluorescence. This method allows for examination under low magnification, enabling faster detection of acid-fast bacteria.

Stain Results
Acid-fast bacteria appear as bright orange-yellow or yellow-green rods with slight curvature, while the background is black. Some residual background material may show a faint yellow color.
