Testing Principle
Under polarized light, collagen fiber tissues exhibit positive uniaxial birefringence. The basic groups in tissue cells adsorb and firmly bind to Sirius Red, enhancing the birefringence and allowing differentiation between different types of collagen fibers.

Stain Results
Under polarized light:

Type I collagen fibers exhibit strong birefringence, appearing bright orange-yellow or vivid red.

Type II collagen fibers exhibit very weak birefringence, appearing light red or bright white, with almost no staining.

Type III collagen fibers exhibit weak birefringence, appearing green.
