Testing Principle
Gram stain was devised empirically by Christian Gram, a Danish physician, in 1884. Gram positive cell wall 
contain thick layer of peptidoglycan with numerous teichoic acid cross linking, after Gram stain, the complex of crystal violet and iodine firmly remained within the cell wall, it is not easy to remove by decolorize solution and continued to show a purple. On this basis, it’s formation of a Gram stain reagents. 
Gram stain is a widely used differential staining method in bacteriology (Belong to counterstain method), can be used for specimen smear or colony smear, after stained bacteria and the environment in stark contrast, you can clearly observe the bacteria morphology, arrangement and some structural features, which used to identify the clinical classification. Staining results will be divided into bacteria Gram-positive bacteria (purple) and Gram-negative bacteria (red) two categories.
