Acid Fast Stain (Kinyoun) (For Auto Stainer) Colourful Clear solution for Medical Diagnose and Medical Research
Testing Principle
Acid-fast bacteria such as Mycobacterium tuberculosis and Mycobacterium leprae are difficult to stain because of their lipoid capsule in the cell wall. However, once the lipoid capsule is stained with an enhanced dye such as carbolfuchsin, the newly formed compound will resist decolorization from acid-alcohol and retain the original color stained (red), which can be easily differentiated from other microorganisms.
This staining kit is based on the Ziehl-Neelsen stain (carbolfuchsin solution) recommended by the WHO.
Mainly used for acid fast bacteria such as mycobacteria of chemical stain. There are two common methods for acid fast stain: 1. Basic fuchsin method also called Ziehl-Neelsen method; 2. Fluorescent dyes method, including Auramine O and Auramine O- Rhodamine B method. This product is used improved basic fuchsin method, the advantage of this method is that it does not need to be heated.

Stain Results
Methylene blue counterstain method:
In light blue background, the acid-fast bacteria are red, and other bacteria and cells are blue.
Malachite green counterstain method:
In light green background, the acid-fast bacteria are red, and other bacteria and cells are green.
Acid fast bacteria (with Mycobacterium tuberculosis as an example): Most Mycobacterium tuberculosis is bacilli and slightly curved. The width of bacteria (0.3~0.6) um, and the length of bacteria is different, between (0.5~8) um, most of them are (1.5~3.5) um. A small number of Mycobacterium tuberculosis may be spiral, and the darker coloring metachromatic granules can be found in the well stained Mycobacterium tuberculosis. Fresh specimens containing more Mycobacterium tuberculosis were observed by acid fast staining and 100x oil microscope. They could not only detect single bacteria, but also cluster or branched arranged bacteria.
