Testing Principle

After staining acid-fast bacteria with the fluorescent dye Auramine O, they are examined under a fluorescence microscope equipped with an ultraviolet light source. When excited by a violet-blue light filter with a wavelength of 350~490 nm, the acid-fast bacteria emit a bright orange-yellow or yellow-green fluorescence. This method allows for observation under low magnification, enabling faster detection of acid-fast bacteria.

Stain Results

Acid-fast bacteria appear as bright orange-yellow or yellow-green, rod-shaped, and slightly curved, against a black background. Some residual materials in the background may show a light yellow color.
