Introduction about incubator
The development of biosimilars—biologic medicines highly similar to an existing approved reference product—requires rigorous analytical comparison. Incubators support several key assays in this characterization.
Cell-based potency assays are central. The biosimilar and reference product are applied to sensitive cell lines (e.g., those proliferating in response to a growth factor) cultured in CO2 incubators. The biological activity (potency) is measured and must be equivalent. Forced degradation studies also use incubators. Samples of both products are stressed under controlled conditions—elevated temperature (e.g., 25°C, 40°C) in stability chambers or incubators—and then analyzed for changes in aggregates, fragments, or charge variants compared to the reference.
Furthermore, incubators are used in assays to detect host cell proteins (HCPs) or in ELISA-based immunogenicity risk assessments. The precision and documentation capabilities of modern incubators are vital here. Any difference in incubation conditions could lead to variable cell responses or degradation profiles, potentially misinterpreted as a product difference. Therefore, the incubator contributes to the “totality of evidence" required by regulators to demonstrate that a biosimilar is as safe and effective as its originator, ensuring patient access to more affordable biologic therapies.
