PROCEDURE
Reagentspreparation:
Allow the reagents to reach room temperature (18-30°C). Check theWash buffer concentratefor thepresenceofsaltcrystals. Ifcrystalshaveformed, resolubilizebywarmingat37°Cuntilcrystals dissolve.DilutetheWashbuffer (20X)asindicatedintheinstructionsforwashing.Usedistilledordeionizedwater andonlycleanvesselstodilutethebuffer.AllotherreagentsareREADYTOUSEASSUPPLIED.
Step1 Preparation:Mark threewellsasNegativecontrol (e.g.B1,C1,D1), twowellsasPositive control (e.g.E1,F1)andoneBlank(e.g.A1,neithersamplesnorHRP-Conjugateshouldbe added into theBlankwell). If theresultswill bedeterminedbyusingdualwavelengthplate reader, the requirement for useof Blankwell couldbeomitted.Useonlynumber of strips

Step3 AddingDiluent:Add100μlofSpecimenDiluent intoeachwellexcept theBlank. AddingSample: Add 10μl of Positivecontrol, Negative control, andSpecimen into their respectivewells except theBlank. Note: Usea separatedisposal pipette tip for each
