Sample Preparation
Solution Preparation
- 10% NaCl (m/v) solution: Dissolve 10g NaCl in 100mL of distilled water
- 20% sugar (m/v) solution: Dissolve 20g sucrose in 100mL of distilled water
- Sample-Diluent: Dilute one volume of 2X Sample-Diluent with one volume of distilled water
Meat Samples (Fish, Shrimp, Beef, Chicken, Pork)
- Homogenize sample with a suitable mixer
- Weigh 0.5g of homogenized sample and mix with 2.5mL of 10% NaCl, vortex vigorously for 3 minutes
- Centrifuge at 4,000 x g for 3 minutes
- Pipette 100μL of supernatant and add 400μL of Sample-Diluent, vortex for 15 seconds
- Use 50μL of supernatant per well for assay

Milk/Plasma/Serum Samples
- Defat high-fat samples by centrifuging
- Pipette 0.5mL of sample and add 4.5mL of 10% NaCl, vortex for 3 minutes
- Centrifuge at 4,000 x g for 3 minutes
- Pipette 100μL of supernatant and add 900μL of Sample-Diluent, vortex for 15 seconds
- Use 50μL per well for assay
Dilution Factor: 100
Milk Powder Samples
- Weigh 0.5g of homogenized sample and mix with 2.5mL of 10% NaCl and 2.5mL of 20% sugar, vortex vigorously for 3 minutes
- Centrifuge at 4,000 x g for 3 minutes
- Pipette 100μL of supernatant and add 400μL of Sample-Diluent, vortex for 15 seconds
- Use 50μL per well for assay
