Pullulanase enzyme PU-730 is prepared from Bacillus licheniformis by submerged fermentation, extraction and purification.
It is so named because it can specifically degrade pullulan polysaccharides (maltotriose, α-1, 6 glycosidic bond-linked polymers).
It belongs to amylase, which specifically cleaves the α-1Power6 glycosidic bond of the branching point of amylopectin and cleaves the whole branched chain structure to form amylose.
PU-730 is usually used in combination with Glucoamylase to produce high glucose syrup and high maltose syrup from liquefied starch.

 

APPLICATION AND BENEFITS

Production of high sugar syrup
Glucoamylase can hydrolyze α-1Magin4 glycosidic bond, but the hydrolysis rate of α-1Magne6 glycosidic bond is low, which is an obstacle to the production of high-sugar syrup.
The combined use of pullulanase and Glucoamylase in the saccharification stage can increase the hydrolysis rate of starch, reduce the amount of Glucoamylase, shorten the reaction time, increase the yield and improve the purity of glucose.

 

Production of ultra-high maltose syrup.
In the production of maltose, the starch is liquefied with α-amylase, and then the starch is decomposed by fungal α-amylase or β-amylase, so the maltose content of the final product is about 50%-60%, which is called high maltose syrup.
Pullulanase combined with α-amylase and β-amylase can produce ultra-high maltose syrup with more than 80% maltose content.

 

Beer brewing.
In beer production, if the starch is not completely decomposed, the content of fermentable sugar will be reduced, and the fermented beer will not meet the requirements.
The synergistic action of pullulanase and α-amylase will make pullulanase completely decompose starch in raw materials, reduce the content of wort in β-limited dextrin, increase the amount of fermentable sugar, and greatly improve the fermentation efficiency.

 

Other applications
The addition of pullulanase in the saccharification process of monosodium glutamate, alcohol, vinegar and soy sauce can increase the glucose content and shorten the saccharification time.
Pullulanase has also found its application in the production of high amylose, which can be further processed into "resistant starch" that is not digested in the small intestine but fermented by intestinal bacteria in the large intestine. this produces short-chain fatty acids that are good for colon health, such as butyrate.
High amylose is also used in adhesive products and the production of corrugated board and paper.
